v cool. The fluorescent agar plate i posted on the prev page is from the Tsien lab and he is far and away the best lecturer i've ever had the privilege of sitting listening agog to. me and about a thousand others. As i said in Oolons thread in E/O its rare to hear a presentation where you could hear a pin drop in a room of a thousand or so people but he had that easy fascination and enthusiasm for his subject that somehow passes over for certain people.

Congrats Prof Tsien.

And heres to fluorescent glowy shit without which i wouldnt have a job. or this job that i love, anyway.

Yes, that one's a beauty. But I'm just astounded by the colour and variety of all those mantids. They don't even look real, but I bet they all disappear when in their own habitat.

Lots of organic compounds exhibit autofluorescence. H&E stains autofluoresce over most of the visible spectrum. I took these on Friday to show somebody why trying to see GFP in a tissue already stained with haematoxylin wasn't such a great idea.

I had to explain to someone today why its wasnt acceptable to carry out a cell death assay when he'd neglected to include a (hopefully) healthy non treated control. And despite me saying hte same thing to him on the last exp. He seems to think that if he is a bit short of cells then it is the best bet to lose all his controls and just do the treated samples, and i have explained to him on multiple occasions that by doing so he invalidates the entire fucking experiment. STILL he doesnt seem to get it. "

I swear some people need to go back to fucking high school.

These I did today just for fun before bleaching the snot outta them with the tornado bleach function on the FV1000



I gotta say, my high resolution bvp cells look like crap after the lossy compression. Is there some way to post the TIF file here?

Thanks, I'll try that format and see how it turns out. It's disappointing to create an awesome image only to have to display it for others in jpeg. I did that one at 1028 x 1028 with a 60X oil /1.42 NA lens. It's a superb lens. Too bad those 2 cells are sitting on top of each other. I had to lower the camera sensitivity a little because where they overlap the pixels were saturated. Consequently the green channel is a little faded looking, especially in this file format. It looked a whole lot better as an oib file on our gigantico hi-res monitor though Oh well.

ETA: Scratch camera replace with PMT...obviously time for me to get some sleep.

yeah i'm glad to say that was a student, one who is incapable of listening unfortunately. His profs on a conference trip at the mo but a quiet word in her shell like when she comes back is probably in order. I honestly dont know how the fuck such people wind up on ANY science course at unniversity, unless they mistakenly think that user of controls is just something that noobs do. this guy is notoriously flakey though anyway and luckily such people are rare, but it amazes me that ANY get through the net.

yeah the compression always makes our images seem pretty dark. I havent found a great way around it other than resizing them tbh.
